Pharmaceutical Solids — The Amorphous Phase
نویسنده
چکیده
“Pharmaceutical Solids” discusses scientific principles associated with pharmaceutical solids useful to practitioners in validation and compliance. We intend this column to help the understanding of principles associated with pharmaceutical solids and to be a useful resource for daily work applications. Enhanced process understanding is an important objective of the quality-by-design initiative. The key objective for this column: Usefulness. Reader comments, questions, and suggestions are needed to help us fulfill our objective for this column. Case studies illustrating principles associated with pharmaceutical solids submitted by readers are most welcome. Please send your comments and suggestions to column coordinator John Bauer at [email protected] or to coordinating editor Susan Haigney at [email protected].

Glass-state amorphous salt solids formed by freeze-drying of amines and hydroxy carboxylic acids: effect of hydrogen-bonding and electrostatic interactions.
We studied effect of molecular interactions on the physical properties of binary freeze-dried solids and frozen aqueous solutions using model chemicals containing various functional groups (amino, carboxyl, hydroxyl). Thermal analysis of frozen solutions containing alkyl diamines and hydroxy di- or tricarboxylic acids showed thermal transitions (T(g)': glass transition of maximally freeze-conce...

متن کاملBuilding solids inside nano-space: from confined amorphous through confined solvate to confined 'metastable' polymorph.
The nanocrystallisation of complex molecules inside mesoporous hosts and control over the resulting structure is a significant challenge. To date the largest organic molecule crystallised inside the nano-pores is a known pharmaceutical intermediate - ROY (259.3 g mol(-1)). In this work we demonstrate smart manipulation of the phase of a larger confined pharmaceutical - indomethacin (IMC, 357.8 ...
متن کاملTheory of amorphous ices.
We derive a phase diagram for amorphous solids and liquid supercooled water and explain why the amorphous solids of water exist in several different forms. Application of large-deviation theory allows us to prepare such phases in computer simulations. Along with nonequilibrium transitions between the ergodic liquid and two distinct amorphous solids, we establish coexistence between these two am...
متن کاملSolid State Crystallinity, Amorphous State, and Its Implications in the Pharmaceutical Process
Many drugs exist in crystalline solid form due to reasons of stability and ease of handling during the various stages of drug development. Crystalline solids can exist in the form of polymorphs, solvates or hydrates. Phase transitions such as polymorph inter-conversion, formation of hydrates, desolvation of solvates, and conversion of the crystalline to amorphous form may occur during various p...
